The Yakuza game series showed me how dense game-worlds can be much better than big empty game-worlds. Rockstar has the capacity to make both a fairly large and dense world happen. I would prefer more interactivity and more interior worlds than just having a huge city with nothing to do. GTA also needs a good offroading area (something for example GTA IV was missing), so I hope they add swamps and mountains around Miami.

Given the climate of what they should parody these days, I’d love for them to include some sort of DC city as the north end part of the map.
